Hello I wanted to see which mouthpiece between the Vandoren V16 6M vs. Vandoren V16 6S+ for alto saxophone. It basically is medium chamber vs. small chamber of the same mouthpiece. How bright is the v16 S+ compared to a Jumbo Java? I want some brightness but not too much so would the S+ fit my needs, or should I stick with the V16 6M medium chamber for a nice overall sound?

It happens that I was looking up the differences in that particular mouthpiece line, and was curious about what the S+ designation meant. I think that you need to try the mouthpieces for yourself to answer the question, however here is what I read about them:

"A medium chamber designed for flexibility, (accuracy in pitch and articulation; the alternate choice in sound. Tip: 196 / Facing: Med LongA7
A small chamber designed for strong projection and accuracy in pitch, articulation, and timbre. A fine choice for a lead player. Tip: 204 / Facing: Med Long"

"The new S+ models feature a brand new chamber design that allows more air than typical small chambers. The result is a sound with more body, depth and color in your sound."

Even the old V16 S is much darker than the Jumbo Java. And the newer S+ has a slightly larger chamber because the S was too much for many players. You can find a discussion here on sotw. So, yes, it is possible the S+ may fit your needs.
